Instructions for Use
BMEM Cardiomyocyte Selection is ready-to-use. Bottles can be used straight from the refrigerator (4 °C) and should not be warmed to RT or 37 °C prior to use.
Directions for Use
BMEM Cardiomyocyte Selection is designed for the purification of human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) following differentiation. Selection is typically
initiated between Day 10 –14 of differentiation, when cardiomyocytes are contracting and non-cardiomyocyte populations remain present. After selection, enriched cardiomyocytes can be maintained long-term in BMEM Cardiomyocyte Maintenance.
- On d10 (d14 the latest) of differentiation, verify your population of hiPSC-CMs with visible spontaneous contractions.
- Aspirate BMEM Cardiomyocyte Maintenance media and replace with BMEM Cardiomyocyte Selection.
- After 48 hours aspirate all media and replace with BMEM Cardiomyocyte Selection.
- After 48 hours aspirate all media and replace with BMEM Cardiomyocyte Maintenance.
Purified populations of hiPSC-CMs should be seen by d14. After selection, BMEM Cardiomyocyte Selection can be aspirated and replaced with BMEM Cardiomyocyte Maintenance. Cardiomyocytes can be maintained in BMEM Cardiomyocyte Maintenance for >45 days.
We recommend replating hiPSC-CMs to new wells for assays between d16 and d20.

Not for diagnostic procedures.
1. Cells do not seem to purify.
Cause: Cardiomyocyte purity was too poor. BMEM Cardiomyocyte Selection works best on cardiomyocytes thatare >50% pure.
- Solution: Complete new differentiation.
2. Cells cease to beat during selection.
Cause: Cardiomyocyte purity was too poor, and only a small population was comprised of contractile cardiomyocytes.
- Solution: Aspirate the selection media and add BMEM Cardiomyocyte Maintenance. If cells don’t start spontaneously beating after 24 h, complete a new differentiation.
